Quantitative monitoring of proteins biomarkers levels in different fluids and tissues by proteomic approaches represents an useful diagnostic and prognostic clinical tool. Methods including data-dependent acquisition are a gold standard for biomarker quantification and validation. The increased sensitivity, resolution and accuracy of mass spectrometers allows precursor ion quantification, which is a good alternative to conventional mass spectrometric methods.
We report a comparision quantification study of glycopeptides from different types of cancers by using both MRM and Ultra-high resolution MS survey scan approach. Selected glycoforms were monitored in albumin depleted human sera by nano liquid chromatography coupled to either ABI Sciex Triple Quad 5500 System or 12T FTICR, solariX XR.
